As shown in the above picture knuckle is attached to dust cover by 4 bolts and to caliper by two bolts. i have only knuckle and not the whole assembly. Should I apply SPC on all these points or it should be left unconstrained?

If you had cover and other components you can use contacts,
Since you don't have other components, use SPCs and carry the analysis.
