If you are doing incompressible both these settings are not required.
Absolute pressure offset would be necessary for compressible cases likes workshop5 compressible nozzle.Nodal Initial conditions for pressure are generally not required. NIC typically help in convergence. Generally I would use them for temperature and eddy viscosity (at times for velocity, it is a flow only in one direction).

So, for incompressible flow what is the means of negative pressure? Does it means that actual pressure is lower than atmospheric pressure (1 atm)?

For incompressible flow, negative pressure would mean lower than atmospheric pressure. (aka negative relative pressure)
Absolute negative pressure would not make sense.0
